Advantages and disadvantages of mammalian cells protein expression

Protein expression in mammalian system has the advantage of generating proteins with folding and post-translational modifications (PTM) identical to native endogenous proteins. In particular, N-glycosylations are correct and end with a sialic acid, and it is therefore a system of choice for the production of glycoproteins. This system is perfectly suited to the production of antibodies, proteins for use as standard in ELISA or mass spectrometry for example.
Expression systems in mammalian cells have also long been favored for the production of therapeutic molecules, but recent innovations revive the interest in yeast systems. Indeed, the major disadvantages of mammalian cells (HEK293 and CHO) are their production yield lower than other eukaryotic systems, their slow growth rate and the higher cost due to the need for complex growth media.
