- Antigen production in E. Coli:
We design particular antigens to have the highest propability to get custom monoclonal antibodies that will detect accessible epitopes. We usually build a PEC –Poly Epitope Complex- a special assembly of up to 14 selected peptides to be produced in E. Coli. All peptides are separated by specific spacers and cloned in a proprietary vector containing a particular proprietary carrier: HIRE technology (High Immunognenicity Response Enabler). Both systems combined allow to dramatically increase immunogenicity, to ensure the best specificity and select the most accessible epitopes to have high affinity custom monoclonal antibodies for Immunohistochemistry (IHC).

- Subcloning of every positive clone :
The main advantage of this is to be able to select the best clones based on final products and not on parental clones that could lead to completely different subclones, maybe not even secreting any antibody…
- Mammalian cell line transfection :
Cloning of the cDNA into an eucaryotic vector and transfection of a cell line to over express the protein.
- Immunocytochemistry Screening :
Selection of all positive clones working in ICC on over expressed cells (produced previously) and on the same normal cells as a negative control. Specificity is controlled using a GFP tag included in the construct and an anti-GFP antibody. This powerful screening method allows us to select clones that have the highest probability of success to specifically detect the endogenous protein in ICC.
- Ascites :
We will then send you 50µl of unpurified ascites of every positive clone in order for you to choose the best one(s) in immunohistochemistry (IHC). If you want more than 1 clone, we will deliver each additional clone at extra charge.
Our Guarantees:
- LEVEL 1:
We guarantee to deliver at least one clone which will work in Immunocytochemistry ICC on the transfected cell line over expressing the protein. In case we fail, we will refund everything except 300€.
- LEVEL 2:
If we meet the level 1 guarantee but you don’t find at least one clone that works in your lab in Immunohistochemistry IHC against the endogenous protein, we will refund you 70% of the total cost.
Deliverables:
- 2 frozen vials of the guaranteed custom monoclonal antibody clone that works in IHC against the native protein.
- 2mg of purified ascites produced from that clone.
Timeline:
120 days for the total package, antigen production and purified antibody production included.
Data provided:
For each cell line:
- Comprehensive validation data on overexpressed cell lysates in all applications : ELISA, WB, IP and IF data.
- Epitope and affinity data (if available)
- ELISA and dot blot data on immunogen
