A) Prokaryotic system
Proteogenix benefits from a great experience in custom recombinant protein production and purification in Escherichia Coli. We offer a large panel of solutions that allows almost always to obtain the material required for example to develop your antibodies. We can also produce in large scale if needed.
Recombinant protein expression tests :
- Transformation into high yield bacteria strain
- Bacterial culture in different conditions (300ml each)
- Synthesis induction
- Affinity purification on a specific resin
- PAGE analysis (lysats)
NB : we perform several runs of tests to determine : 1) if the protein is soluble or accumulates in inclusion bodies. 2) the production yield. At the end of this step the customer is informed of the feasibility. Along with our advice the customer will decide to stop or go to the next step and then define the amount to produce.
Production and purification of the recombinant protein :
- Bacterial culture: 1 litre
- Synthesis induction according to the protocol established above
- Bacterial lysates preparation
- Affinity purification using a specific resin
- Option : elimination of the tag via enzymatic cleavage (GST tag)
NB : This step is performed according to the decision made by the customer after the second step. Proteogenix cannot guarantee the final amount of protein obtained after the scale-up (performed with 0.3 to 1 litre or more).
Expected duration
-Expression tests : 10 to 20 days
-Production and Purification : 5 to 10 days
